Mavyret (glecaprevir/pibrentasvir) is an oral combination antiviral medication made by AbbVie for the treatment of chronic hepatitis C virus (HCV) infection. It combines two direct-acting antivirals (DAAs) — glecaprevir (an NS3/4A protease inhibitor) and pibrentasvir (an NS5A inhibitor) — into a once-daily pill that can cure hepatitis C in as few as 8 weeks for most patients.

What Does Mavyret Treat?

Mavyret is FDA-approved to treat:

How Mavyret Works

HCV replicates inside liver cells using viral proteins — NS3/4A protease and NS5A — that are essential for processing viral proteins and replicating the genome. Glecaprevir blocks NS3/4A protease, which is required to cleave the HCV polyprotein into functional viral proteins; pibrentasvir blocks NS5A, a protein involved in RNA replication and viral assembly. By targeting two separate stages in the HCV lifecycle simultaneously, Mavyret achieves sustained virologic response (SVR) rates above 97% across all genotypes — meaning the virus is undetectable 12 weeks after completing treatment, which is considered a functional cure.
