Venclexta (venetoclax) is a targeted oral cancer therapy co-developed by AbbVie and Genentech (Roche). It belongs to a class called BCL-2 inhibitors — the first drugs that work by blocking BCL-2, a protein that prevents cancer cells from self-destructing, restoring the cell's natural ability to undergo programmed cell death.

What Does Venclexta Treat?

Venclexta is FDA-approved for:

  • Chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma (SLL) — as a first-line treatment (combined with obinutuzumab) or after at least one prior therapy (combined with rituximab or as monotherapy)
  • Acute myeloid leukemia (AML) — in combination with azacitidine, decitabine, or low-dose cytarabine, for newly diagnosed adults 75 and older or who have comorbidities that prevent intensive chemotherapy

How Venclexta Works

BCL-2 is an anti-apoptotic protein that many cancer cells overexpress to survive — it blocks the cell's internal "self-destruct" signals. Venetoclax is a selective, potent BCL-2 inhibitor that binds directly to the BCL-2 protein and prevents it from blocking apoptosis (programmed cell death). This restores the cancer cell's ability to self-destruct, causing rapid tumor cell death — a mechanism called "direct tumor lysis." Because venetoclax works through a fundamentally different pathway than traditional chemotherapy, it can be effective even in patients whose cancer has become resistant to prior treatments.
